Physicochemical characterization of mitochondrial DNA from soybean.
Mitochondrial DNA (mtDNA) of soybean (Glycine max L.) was isolated and its buoyant density was contrasted with that of nuclear (nDNA) and chloroplast (ctDNA) DNA. Each of the three DNAs banded at a single, characteristic buoyant density when centrifuged to equilibrium in a CsCl gradient. Buoyant densities were 1.694 g/cm(3) for nDNA and 1.706 g/cm(3) for mtDNA. These values correspond to G-C co...
متن کاملPhysicochemical characterization of mitochondrial DNA from pea leaves.
The mitochondrial DNA from pea leaves exists in a circular conformation. 25% of the circular molecules exist as supercoils, and 10% of the molecules are dimers. The molecular weight of mitochondrial DNA is about 66 to 70 x 10(6) by electron microscopy, and 74 x 10(6) from its renaturation kinetics. No evidence for inter- and intramolecular heterogeneity is found.
متن کاملIsolation and Characterization of Mitochondrial Dna from Drosophila Melanogaster
Mitochondrial DNA (MtDNA) with a neutral buoyant density of 1.681 g/cm(3) has been isolated from unfertilized eggs of Drosophila melanogaster. This DNA is a circular molecule with an average length of 5.3 microm; it reassociates with a low C(0)t(1/2) after denaturation, and in alkaline isopycnic centrifugation it separates into strands differing in density by 0.005 g/cm(3). MtDNA isolated from ...
متن کاملCharacterization of mitochondrial DNA from the pika (Ochotona rufescens rufescens).
Further to anatomical and physiological studies performed on the pika (Ochotona rufescens rufescens), a new laboratory animal, the main characteristics of the mitochondrial DNA from its liver are defined. The buoyant density of this DNA is 1.695 g/cm3, its length 5.30 mum,, i.e., 3.17 times that of the replicative form of phiX 174. It could have approximately 16 500 base pairs. The DNA from the...
متن کاملIsolation and characterization of mitochondrial DNA from Chlamydomonas reinhardtii.
Mitochondrial DNA (mtDNA) has been isolated from a mitochondrial pellet of Chlamydomonas reinhardtii. The mtDNA has a buoyant density of 1.706 g/ml in CsCl, a melting temperature of 87.9 degrees in standard saline citrate, and a nucleoside composition of 47.5% deoxyguanidine plus deoxycytidine with no odd nucleosides. Thermal denaturation and renaturation studies have shown that (i) mtDNA conta...
